鳄嘴花的组织培养和快速繁殖

摘 要:

以鳄嘴花[Clinacanthus nutans (Burm. f.) Lindau]幼嫩茎段为外植体, 研究不同植物生长调节剂对鳄嘴花茎段腋芽诱导、丛生芽分化、增殖及生根培养的影响。结果表明: 茎段在培养基MS+1.0 mg·L-1 BAP+0.1 mg·L-1 NAA上诱导产生腋芽, 将腋芽转入培养基MS+1.0 mg·L-1 BAP+0.1 mg·L-1 NAA诱导分化丛生芽, 分化率高达93.3%; 在培养基中分别加入30 g·L-1椰汁、30 g·L-1香蕉、0.5 g·L-1蛋白胨, 都有壮苗的效果; 最佳的生根培养基为MS+0.5 mg·L-1 NAA+2.0 mg·L-1 IBA, 生根率达90%, 且根系发达, 芽苗生长健壮。移栽至混合基质(泥炭:椰糠:珍珠岩:河沙=3:2:2:1)中, 鳄嘴花的成活率达97%。

Tissue culture and rapid propagation in vitro of Clinacanthus nutans

Abstract:

The regeneration system of tissue culture of Clinacanthus nutans was established by utilizing its in vitro-derived stem as explant. We analyzed the differentiations characteristics of explant, and discussed affecting factors of tissue culture of hormone and additional nutrient. The results showed the optimal proliferation medium for C. nutans was MS with 1.0 mg L-1 BAP and 0.1 mg·L-1 NAA. The plantlets grew well when the organics was adding, 0.5 g·L-1 peptone, 30 g·L-1 coconut juice and 30 g·L-1 banana were good for cultivating strong seedling. A rooting medium composed of MS with 0.5 mg·L-1 NAA and 2.0 mg·L-1 IBA provided 90% rooting and vigourous plantlets. Plantlets were transplanted to a potting mixture (3:2:2:1, peat: coconut coir: perlite: sand) in trays with about 97% survival percentage.
